What it is

Mono- and diglycerides are glycerol bound to one or two fatty acids; ordinary fat has three 12.

E471 (INS 471), an emulsifier, is a mixture of glycerol mono-, di- and triesters of fatty acids, usually made by glycerolysis of fats and oils, which may be hydrogenated 341. It is used mainly in desserts, ice cream, bread and margarine 1.

How often it is used

E471 is the tenth most frequent additive in French packaged foods, listed in 6,060 of 126,556 products in 2019 5.

Of 106,489 adults, 78.07% consumed it, at a cohort mean of 160.8 mg/day 6.

In the body

Digestive enzymes very likely split E471 into glycerol and fatty acids 1.

Enterocytes re-esterify absorbed fatty acids to triacylglycerol, mostly by the monoacylglycerol pathway 2. EFSA’s mean adult intake estimates equal 0.8 to 3.5% of reference fat intake 1.

In one donor’s faecal cultures, several measures were altered irreversibly by glyceryl stearate and one or none by two other E471 products 7.

What studies show

In a French cohort, higher E471 intake was linked to slightly more cancer 8; see also emulsifiers.

Among 92,000 adults over a mean 6.7 years, hazard ratios, highest against lowest third of intake, were 1.15 (95% CI 1.04 to 1.27) overall, 1.24 (1.03 to 1.51) for breast and 1.46 (1.09 to 1.97) for prostate cancer; modelled absolute risk at age 60 was 12.4% against 11.2% 8. E471 was not associated with type 2 diabetes over a mean 6.8 years 9.

EFSA judged a numerical acceptable daily intake unnecessary; JECFA set one “not limited” in 1973 1.

Above 200 °C, diglycerides form glycidyl esters, hydrolysed in the gut to glycidol, a genotoxic carcinogen in rodents 110.

Regulation

Four jurisdictions permit E471 113144.
